How CLOFARABINE Works
Clofarabine is intracellularly metabolized to its active 5’-triphosphate metabolite, which inhibits DNA synthesis through two primary mechanisms: the reduction of cellular deoxynucleotide triphosphate pools via inhibition of ribonucleotide reductase, and the termination of DNA chain elongation through competitive inhibition of DNA polymerases. Additionally, the drug disrupts mitochondrial membrane integrity, triggering the release of pro-apoptotic factors (cytochrome C and apoptosis-inducing factor) to induce programmed cell death in both proliferating and quiescent cancer cells.
