Pioglitazone hydrochloride and glimepiride is a combination medication indicated as an adjunct to diet and exercise to improve glycemic control in adults with type 2 diabetes mellitus. This combination therapy is not recommended for the treatment of type 1 diabetes or diabetic ketoacidosis. It combines two antihyperglycemic agents with complementary mechanisms of action to manage blood glucose levels.

How PIOGLITAZONE HYDROCHLORIDE AND GLIMEPIRIDE Works
This combination product functions through two distinct mechanisms. Glimepiride, a sulfonylurea, stimulates insulin release from pancreatic beta cells by binding to sulfonylurea receptors and closing ATP-sensitive potassium channels. Pioglitazone, a thiazolidinedione, is a potent and selective agonist for peroxisome proliferator-activated receptor-gamma (PPARγ). It improves glycemic control by increasing insulin sensitivity in muscle and adipose tissue and reducing hepatic glucose production. Together, these agents increase insulin secretion and decrease insulin resistance.
