Understanding FLT3
What it means: FLT3 mutations (ITD insertions, TKD point mutations) cause constitutive kinase activation. Found in ~30% of AML patients.
Why it matters: FLT3 inhibitors (midostaurin, gilteritinib, quizartinib) improve outcomes when added to chemotherapy or as monotherapy.
Testing: Tested via PCR (ITD) and sequencing (TKD). Allelic ratio of ITD affects prognosis. Tested at diagnosis and relapse.
