Gralise is indicated for the management of postherpetic neuralgia (PHN). It is not interchangeable with other gabapentin products due to its unique pharmacokinetic profile, which dictates a specific dosing frequency. This medication is specifically used to manage neuropathic pain associated with the aftermath of herpes zoster infections.

How GRALISE Works

The exact mechanism by which gabapentin exerts its analgesic effect is unknown. In animal models of neuropathic pain, it prevents allodynia (pain from non-painful stimuli) and hyperalgesia (exaggerated pain response). While gabapentin is a structural derivative of the neurotransmitter GABA, it does not bind to GABA receptors, is not converted into GABA, and does not inhibit GABA uptake or degradation. It also shows no affinity for other common receptor systems, including benzodiazepine, glutamate, NMDA, dopamine, or serotonin receptors. Research indicates it binds with high affinity to the alpha-2-delta subunit of voltage-gated calcium channels.
