How PEMETREXED DISODIUM Works
Pemetrexed disrupts folate-dependent metabolic processes essential for cell replication. It enters cells via the reduced folate carrier and is converted into active polyglutamate forms by the enzyme folylpolyglutamate synthetase. These polyglutamated metabolites are retained within cells and inhibit three key enzymes in the de novo biosynthesis of thymidine and purine nucleotides: thymidylate synthase (TS), dihydrofolate reductase (DHFR), and glycinamide ribonucleotide formyltransferase (GARFT). Polyglutamation is more extensive in tumor cells than normal tissues, resulting in prolonged drug action in malignant cells.
