Gefitinib is a tyrosine kinase inhibitor indicated for the first-line treatment of patients with metastatic non-small cell lung cancer (NSCLC) whose tumors harbor epidermal growth factor receptor (EGFR) exon 19 deletions or exon 21 (L858R) substitution mutations, as detected by an FDA-approved test. Its efficacy has not been established in patients with EGFR mutations outside of these specific markers.

How GEFITINIB Works

Gefitinib is a reversible inhibitor of the EGFR tyrosine kinase. It binds to the intracellular catalytic domain of the receptor, preventing the autophosphorylation of tyrosine residues. This action blocks downstream signaling pathways that drive tumor cell proliferation, survival, and metastasis. Gefitinib demonstrates a higher binding affinity for EGFR exon 19 deletions and exon 21 (L858R) mutations compared to wild-type EGFR. It also inhibits IGF and PDGF-mediated signaling at clinically relevant concentrations.
